PocH-100i is a small, single unit haematology analyser capable ofgenerating a CBC with 3-part DLC using only 2 reagents. After manual mixingthe sample has to be loaded to the instrument. In combination withcorresponding adaptors a whole variety of tubes can be used. The requiredsample volume is 1 ml in a standard collection tube (500 [micro]l in amicrotube). Sample number registration can be done manually or by a hand holdbar code reader. The instrument has a throughput around 25 samples per hour.In brief, haemoglobin (Hb) is determined by a noncyanide method; red bloodcells (RBC), white blood cells (WBC) and platelets (PLT) are detected bydirect current (DC). MCV is measured from the RBC electrical pulse analysis.Other CBC results (haematocrit, MCH, MCHC, Red cell Distribution With) arecalculated. A 3-differential is obtained by separating the WBC histogram into3 areas using 4 discriminators (lower discriminator, T1, T2 and upperdiscriminator). Lymphocytes are located between the lower and T1discriminator; monocytes, eosinophils and basophils are located between theT1 and T2 discriminator and designated together as "mixed cells";neutrophils are located above the T2 discriminator. Results (includingquality control data) can be printed using the integrated printer and can betransmitted to a host computer. A more extensive description of theinstrument as well as its comprehensive flagging system have been describedelsewhere (2). The manufacturer's specifications for linearity are: WBC[1.0-99.9] x [10.sup.9]/l, RBC [0.3-7.0] x [10.sup.12]/l, platelets[10.0-999.0] x [10.sup.9]/l and Hb [0.1-25.0]g/dl. Apart from a dailystart-up and shut-down procedure, maintenance included a daily "cleantransducer" cycle and a weekly "waste chamber cleaning" cycle.Daily and weekly maintenance are performed within 20 and 25 minutesrespectively.

For the comparison studies a large series of oncology samplescovering both values in the normal clinically range and values in thecritically low range, frequently encountered in an oncology setting wereincluded. As recommended by the guidelines, analyses were performed by atrained non-laboratory user and an experienced laboratory technician (3).Despite the different technology the poch-100i showed excellent correlationwith the CD4000 instrument. The pocH-100i also had excellent correlation withthe XE-2100 instrument using similar technology. Comparison of subgroups forplatelets with results in the lower range gave lower correlation([r.sup.2]=0.82). A difference in measuring principle between instruments ispartly an explanation but it is well known that even identical instrumentsshow a weaker correlation in the lower platelet range (8,11). The results ofour correlation studies are comparable to other published reports (2).


PocH-100i vs Manual Differential The WBC from the ABX Pentra 80 was used to calculate the absolute neutrophil counts from the manual differentials. The instruments showed excellent correlation (R2 = 0.9779) where there were no flags but correlation was not as good where flags were seen (R2 = 0.9050).


As expected, correlation where instrument flags were present was not as good however the values were still acceptable with the worse correlation between the PocH-100i and manual differential and between the PocH-100i and the ABX Micros ES 60. The best correlation, unsurprisingly, was seen between the five part differential analyser and manual differential but the ABX Micros ES 60 also compared well with both manual and automatic 5-part differentials. 041b061a72
